Shiitake Mushroom Fruiting Body Extract

Also known as: Lentinula edodes, Shiitake mushroom, Black forest mushroom, Oak mushroom

Overview

Shiitake mushrooms (*Lentinula edodes*) are native to East Asia and cultivated globally for culinary and medicinal uses. Rich in nutrients like proteins, carbohydrates, vitamins, and minerals, they are valued for potential health benefits. The fruiting bodies and mycelium contain bioactive compounds such as lentinan (a beta-D-glucan polysaccharide), eritadenine, and other polysaccharides and lignans. Shiitake mushrooms are used for their potential anticancer, immunostimulatory, antiviral, hepatoprotective, and antihypercholesterolemic effects. Research is moderately advanced, with numerous preclinical and some clinical studies suggesting efficacy, though more rigorous human trials are needed to confirm findings. They are available in various forms, including whole dried mushrooms and mycelial extracts.

Benefits

Shiitake mushrooms offer several potential health benefits. They significantly improve immune function by increasing the proliferation of γδ-T cells (60% increase, p < 0.0001) and NK-T cells (2-fold increase, p < 0.0001), and enhancing secretory immunoglobulin A (sIgA) production. Lentinan, a polysaccharide from shiitake, has demonstrated antitumor activity, with a meta-analysis showing a modest increase in overall survival (OS) among patients with advanced gastric cancer treated with lentinan plus chemotherapy (hazard ratio for OS, 0.8; 95% CI, 0.68-0.95). Additionally, shiitake may support cardiovascular health by lowering serum cholesterol levels and preventing dyslipidemia, primarily through the compound eritadenine. They also exhibit hepatoprotective effects, improving liver function and reducing liver inflammation markers in animal studies. Shiitake mycelial extract has been found to decrease chemotherapy-associated adverse effects and improve quality of life (QOL) in patients with advanced gastrointestinal cancer.

How it works

Shiitake mushrooms exert their effects through several biological pathways. They stimulate the immune system by enhancing the proliferation and activation of γδ-T cells and NK-T cells, and increasing the production of cytokines such as IL-4, IL-10, and TNF-α. Lentinan exhibits antitumor effects by suppressing cytochrome P450 1A enzymes, inducing apoptosis in tumor cells, and increasing serum IL-2 and TNF-α levels. Eritadenine regulates lipid metabolism by inhibiting S-adenosyl homocysteine hydrolase activity, which helps in reducing cholesterol levels. These mechanisms collectively contribute to the observed health benefits.

Side effects

Shiitake mushrooms are generally safe when consumed in moderate amounts. Common side effects include mild diarrhea and skin rash, which typically resolve as the body adapts. Uncommon side reactions may include a slight increase in liver enzymes (GOT and GPT) and a feeling of mild pressure on the chest. Rare side effects are not significantly reported in the literature. Shiitake extracts may interact with blood thinners by decreasing the effectiveness of blood platelets in initiating coagulation. Individuals with allergies to mushrooms or those prone to "mushroom worker's lung" should avoid shiitake supplements. Pregnant or breastfeeding women should consult healthcare providers before use due to limited safety data.

Dosage

While the minimum effective dose is not well established, studies have used doses ranging from 5 to 10 grams of dried shiitake mushrooms per day, which has shown beneficial effects on immune function and other health parameters. There is no established maximum safe dose, but high doses (over 50 mg/day of mycelium extract) have been used without significant toxicity in animal studies. Consistent daily consumption is typically used in studies, but no specific timing recommendations are available. Both whole dried mushrooms and mycelial extracts have been used, with the latter often being more concentrated in bioactive compounds. The bioavailability of shiitake compounds can vary depending on the preparation method and individual digestive health.
